Tranmere will provide a stern test with them seeing off Stockport County in the first round 4-1, while the Railwaymen beat AFC Fylde 4-0.
Kick-off for the 2nd round tie is 7pm at Prenton Park with prices set at £3 for adults and £1 for concessions.
The winners will take on Bolton Wanderers in mid December at the Reebok Stadiium.
